v twins can be weird, what is the crank trigger as in how many teeth and gaps?
I just had a Suzuki V twin apart and the cylinders may be 75 deg apart but the rods don't share the same pin so it doesnt fire 75 deg apart

Yes 40 or 45 deg would be a good start. I had to use pot to start and stop cut, this is the only option when you are using the S100 as a sequential gearbox controller, it works fine. Ignition Retard is kinder on the engine, use retard as a preference.
You might want to try pot to start and stop the cut dependant on how much slop there is in the selector barrel.
Sorry not sure about the load cell.

Can you use the laptop to confirm what com port it thinks it has, somewhere in device manager or such like this will be found. Then set the DTA software to use that port and then try to connect, this can be found at File/Options

If you intend to test sensor position on the starter motor, you will need to set 'firing tooth on start up' to '0' this should be close to TDC, its harder to explain than do! Yes that's it, once running use a timing light to verify position. I get confused easy when using a wasted spark and timing light (I assume you are not using a distributer for sparks, and are using a wasted spark setup) so I enter 0's into the ignition table and look for the alignment of the sparks ...

Reading some of your older posts I note that this engine is a 6 cylinder twin plug with coil on plug's with built in ignitors. How have you wired this to the ECU?

I think you need to tick 'use left driven wheel speed' and copy the physical undriven input to pin 31 or I think thats how I made it work
